How our peptides are manufactured and released

Sequences are produced by solid-phase peptide synthesis (SPPS), cleaved, purified by preparative reverse-phase chromatography and then lyophilized under vacuum to a stable powder cake. Lyophilization removes the free water that drives hydrolysis and deamidation, which is why correctly stored lyophilized peptide is far more stable than a peptide in solution.

Before a lot is released it is assayed in house by RP-HPLC for purity and then submitted to an independent analytical laboratory for confirmatory HPLC and mass spectrometry. The independent report establishes both purity (area percent at the primary peak) and identity (observed mass against theoretical mass). Both are published on the certificate for that lot.

Reading a certificate of analysis

A usable COA states the lot number, the analytical method, the purity figure with its integration window, the observed molecular mass, and the date of test. Purity expressed without a method is not a meaningful figure — a peptide reported at 99% by an unnamed method tells you nothing about what was integrated or excluded.

Where a peptide is supplied as an acetate or trifluoroacetate salt, the net peptide content differs from the gross vial weight. Laboratories performing quantitative work should calculate against net peptide content rather than the labelled milligram figure. Raw chromatograms and spectra for any PrimeGen lot can be requested from laboratory support.

Storage, reconstitution and stability

Sealed lyophilized vials are stored at -20 °C, desiccated and protected from light. Material remains ambient-stable through normal transit windows and ships at room temperature, so a vial that arrives warm has not been compromised — return it to freezer storage on receipt.

Reconstitution parameters depend on the protocol. Diluent is added slowly against the vial wall and the vial is swirled rather than shaken, because mechanical shear and foaming both degrade peptide. Once in solution, peptides are held at 2–8 °C and used within a short working window; repeated freeze-thaw cycling is the most common avoidable cause of assay drift.
