Research background

CJC-1295 without DAC, commonly written as Modified GRF (1-29), is the 29-residue GHRH fragment carrying four stabilising amino-acid substitutions but no albumin-binding linker. It is the short-acting counterpart used where pulsatile receptor stimulation must be preserved.

Mechanism of action

Binds the GHRH receptor and activates Gs-coupled cAMP signalling in pituitary somatotrophs. The substitutions at positions 2, 8, 15 and 27 resist DPP-4 cleavage and trypsin-like degradation relative to native GHRH, while clearance remains fast enough to model physiological pulses.

Reconstitution

Allow the vial to reach room temperature before opening to prevent condensation on the lyophilized cake. Add sterile or bacteriostatic water slowly down the inner wall of the vial, then let the powder dissolve without agitation; swirl gently if needed. Do not shake or vortex — shear forces denature peptide secondary structure and cause foaming that traps material on the stopper.

Stability

As a sealed lyophilized powder the material is stable for extended periods at -20 °C when kept desiccated and protected from light. Once in solution, stability shortens considerably: aqueous stocks are normally held at 2-8 °C for short term work or aliquoted and frozen for longer storage. Repeated freeze-thaw cycling is the most common cause of measurable potency loss.
