Research background

GHK-Cu is the copper(II) complex of the tripeptide glycyl-L-histidyl-L-lysine, first isolated from human plasma. It is a long-standing model system in matrix-biology research because it couples a defined peptide to a redox-active metal centre.

Mechanism of action

The tripeptide has high affinity for Cu(II) and acts as a physiological copper shuttle. In dermal fibroblast culture the complex is associated with modulation of collagen and glycosaminoglycan synthesis, metalloproteinase and TIMP balance, and broad transcriptional shifts consistent with copper-dependent enzyme activity.

Reconstitution

The copper complex gives a characteristic deep blue solution on reconstitution; colour intensity is not a purity indicator. Avoid chelating buffers such as EDTA, which strip the copper centre.

Stability

As a sealed lyophilized powder the material is stable for extended periods at -20 °C when kept desiccated and protected from light. Once in solution, stability shortens considerably: aqueous stocks are normally held at 2-8 °C for short term work or aliquoted and frozen for longer storage. Repeated freeze-thaw cycling is the most common cause of measurable potency loss.
