Melanotan II is a cyclic lactam analog of alpha-MSH with broad activity across melanocortin receptor subtypes, while PT-141 (bremelanotide) is its metabolite with a comparatively MC4R-weighted profile. Because they are structurally close, receptor-subtype selectivity — not simple binding affinity — is the meaningful distinction in research design.
Cyclic peptides in this family are conformationally constrained, which makes them chromatographically distinctive and generally more stable than their linear counterparts.
Semax and selank are short synthetic sequences derived respectively from ACTH(4-10) and tuftsin, studied in neurotrophic-factor expression and neurochemical models. Both are short, highly soluble and early-eluting. Dihexa is a very different construct — an angiotensin IV analog studied in hepatocyte-growth-factor and synaptogenesis models, and considerably more lipophilic.
Solubility varies sharply across this group. Lipophilic constructs may require a co-solvent step before aqueous dilution, and the co-solvent then has to be accounted for as a vehicle control in any cell-based assay.
Several compounds in this category are photosensitive; amber vials or foil-wrapped storage are standard once material is in solution. Cyclic and constrained peptides can also show unexpected retention behaviour, so purity methods are developed per sequence rather than applied generically.
