FOXO4-DRI is a retro-inverso peptide built from D-amino acids in reversed sequence order, engineered to mimic the FOXO4 interaction surface while resisting proteolysis. It is a standard tool compound in senolytic research.
Designed to interfere with the FOXO4-p53 protein-protein interaction that sequesters p53 in the nucleus of senescent cells. Disruption is reported to permit p53 mitochondrial translocation and selective apoptosis of senescent, but not proliferating, cells in culture.

Supplied lyophilized; typically reconstituted in sterile water or PBS. As a D-amino-acid construct it is markedly protease-resistant, which simplifies long-incubation culture work.
As a sealed lyophilized powder the material is stable for extended periods at -20 °C when kept desiccated and protected from light. Once in solution, stability shortens considerably: aqueous stocks are normally held at 2-8 °C for short term work or aliquoted and frozen for longer storage. Repeated freeze-thaw cycling is the most common cause of measurable potency loss.
