IGF-1 LR3 is a 83-residue recombinant analog of insulin-like growth factor 1, extended by a 13-residue N-terminal sequence and substituted at position 3 (Arg for Glu). Both modifications sharply reduce binding to IGF binding proteins.
Agonist at the IGF-1 receptor, a receptor tyrosine kinase signalling through IRS-1 to the PI3K-Akt and MAPK cascades. Because IGFBP affinity is reduced by roughly two orders of magnitude, free-fraction concentration in culture media is far higher than for native IGF-1, which is the reason it is preferred in cell-culture supplementation research.
