The practical research consequence is that each requires a different receptor panel to characterise properly.

Cagrilintide sits in a separate family entirely — it is a long-acting amylin analog acting at calcitonin and amylin receptor complexes, not at incretin receptors. Combination research material such as CagriSema pairs an amylin analog with a GLP-1 agonist, so identity confirmation for these blends requires resolving two distinct masses.

Why these sequences need careful handling

Long-acting incretin analogs are typically fatty-acid acylated to promote albumin binding. That lipophilic side chain changes their chromatographic behaviour and makes them more prone to adsorption onto glass and plastic surfaces at low concentration — a common and under-recognised source of apparent potency loss in dilute working solutions.

These peptides are also sensitive to surfactant-free vortexing. Foaming denatures the helical region and produces aggregates that show as shoulder peaks on HPLC. Swirl to dissolve; never shake.

Analytical considerations

Acylated peptides elute late on C18 and can tail if the mobile phase modifier is insufficient. Method development for this class usually needs a wider gradient window than for small unmodified sequences such as BPC-157.
