Melanotan II is a synthetic cyclic analog of alpha-MSH used as a reference ligand in melanocortin receptor research, particularly around pigmentation pathway signalling.

Analytical notes for pigmentation and matrix work

Melanotan II is a cyclic peptide and its ring structure is the basis of its receptor affinity; harsh reconstitution, elevated temperature and extremes of pH can all compromise it. Add diluent gently, avoid shaking, and keep the reconstituted vial cold and dark.

GHK-Cu's copper(II) centre produces a blue solution on reconstitution and can interfere with colorimetric and fluorescence readouts. Where that matters, run matrix and pigmentation arms as separate assays rather than in a shared well plate, and avoid chelating buffers on the GHK-Cu arm.

Lyophilized material is ambient-stable across normal transit windows, so a kit that arrives at room temperature has not been compromised — move it to freezer storage on receipt and record the date.

Reconstitute against the vial wall with bacteriostatic or sterile diluent, swirl rather than shake, and hold reconstituted solution at 2–8 °C for a short working window. Mechanical shear, foaming and repeated freeze-thaw cycling are the three most common avoidable causes of drift between replicate runs.

Each lot in a kit is assayed in house by RP-HPLC and re-tested by an independent laboratory using HPLC and mass spectrometry. Purity is reported as area percent at the primary peak; identity is confirmed by observed mass against theoretical mass. Lot-matched certificates are published digitally and are not enclosed with the shipment.

Why is GHK-Cu supplied at 50 mg when the other vial is 10 mg?
Matrix and fibroblast work typically consumes far more material across a dilution series than receptor-binding work does, so the kit is weighted toward the GHK-Cu arm.
Can both compounds be run in the same assay?
It is usually better not to. Free copper from the GHK-Cu arm interferes with several colorimetric and fluorescence readouts, so most laboratories run the matrix and pigmentation arms separately.
